Robert, a Harvard University symbologist, is summoned to Vatican City to investigate the murder of a cardinal and the theft of a powerful antimatter canister by the Illuminati, an ancient secret society. Langdon is joined by Vittoria Vetra (Ayelet Zurer), a nuclear physicist who is also investigating the theft.

The Illuminati leave a series of clues inspired by the works of Galileo Galilei, a 17th-century scientist who was persecuted by the Catholic Church for his support of heliocentrism. Langdon and Vetra must follow these clues to stop the Illuminati from destroying Vatican City with the antimatter canister.

Along the way, Langdon and Vetra must also contend with the Vatican's own secret police, the Camerlegengo (Ewan McGregor), who is determined to protect the Church at all costs.

In the end, Langdon and Vetra are able to stop the Illuminati and save Vatican City. However, they also learn that the Camerlegengo was the true mastermind behind the plot. The Camerlegengo had planned to use the antimatter canister to destroy Vatican City and bring about a new era of Catholic faith.
